Несмотря на распространенность и частоту применения, вопросы стандартизации методов определения цитотоксичности материалов для медицинских изделий остаются нерешенными. Широкая вариативность подходов к оценке цитотоксичности in vitro требует от исследователя тщательного планирования испытания с учетом особенностей исследуемого объекта.</p></sec><sec><title>Цель</title><p>Цель. Провести сравнительный анализ чувствительности разных культур клеток и методов оценки цитотоксичности при анализе материала для создания протезов для сердечно-сосудистой системы на примере консервированного ксеноперикарда.</p></sec><sec><title>Материалы и методы</title><p>Материалы и методы. Эксперимент проведен на трех культурах клеток: фибробласты, HUVEC, Ea.hy926, с использованием специфичной для каждой культуры питательной среды. В качестве объекта с цитотоксическим действием использовали образцы ксеноперикарда, хранящиеся в растворе парабенов. Анализировали реакцию клеток в прямом и непрямом контакте с образцами ксеноперикарда, а также в присутствии его экстракта. Проводили МТТ-тест, исследование динамики роста клеток с помощью клеточного анализатора xCelligence, оценку пролиферативной активности клеток с помощью коммерческого набора Click-IT.</p></sec><sec><title>Результаты</title><p>Результаты. По результатам проведенного МТТ-теста во всех случаях присутствие в культуральной среде экстракта ксеноперикарда приводило к выраженному снижению жизнеспособности клеток и плотности популяции. Согласно оценке динамики роста, в группах с образцами перикарда отмечали полную гибель клеток. Добавление экстракта консервированного ксеноперикарда не оказало заметного влияния на изменение клеточного индекса роста культуры фибробластов, однако в присутствии образцов ксеноперикарда происходило его выраженное снижение. В культуре HUVEC добавление как экстракта, так и образцов ксеноперикарда в короткие сроки вызывало гибель значительной части клеток. В культуре Ea.hy926 отмечалось снижение темпов пролиферации, однако в течение всего эксперимента продолжалась положительная динамика роста культуры клеток. В присутствии экстракта отмечали отсутствие пролиферирующих клеток в культуре фибробластов, уменьшение их количества в культуре Ea.hy926.</p></sec><sec><title>Заключение</title><p>Заключение. Условия проведения эксперимента, тестовая культура и методы оценки должны подбираться индивидуально исходя из особенностей тестируемого материала. Для получения достоверных результатов необходимо использование нескольких принципиально разных методов исследования для более полной характеристики действия цитотоксических агентов на культуру клеток.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>Highlights</title><p>Highlights</p></sec><sec><title> </title><p> </p></sec><sec><title>Resume</title><p>Resume</p></sec><sec><title>Background</title><p>Background. Despite the prevalence and frequency of use, the issues of standardization of methods for determining the cytotoxicity of materials for medical devices remain unresolved. The wide variability of approaches to assessing cytotoxicity in vitro requires the researcher to carefully plan the test to according with characteristics of the object being studied.</p></sec><sec><title>Aim</title><p>Aim. To conduct a comparative analysis of the sensitivity of different cell cultures and methods for assessing cytotoxicity materials for creating cardiovascular prostheses using preserved xenopericardium as an example.</p></sec><sec><title>Methods</title><p>Methods. The experiment was carried out on three cell cultures: fibroblasts, HUVEC, Ea.hy926, with the specific culture medium for each case. We used xenopericardium samples stored in a paraben solution as an object with a cytotoxic effect. We analyzed reaction of cells in direct and indirect contact with xenopericardium samples, as well as in the presence of its extract. An MTT test was carried out, cell growth dynamics were studied using the xCelligence cell analyzer and cell proliferative activity was assessed using a commercial Click-IT kit.</p></sec><sec><title>Results</title><p>Results. According to the results of the MTT test, in all cases the presence of xenopericardium extract in the culture medium led to a pronounced decrease in cell viability and population density. According to the assessment of growth dynamics, in groups with pericardial samples, complete cell death was noted. The addition of preserved xenopericardium extract did not have a noticeable effect on the change in the cellular growth index of the fibroblast culture, however, in the presence of xenopericardium samples, there was a pronounced decrease in it. In HUVEC culture, the addition of both the extract and xenopericardium samples caused the death of a significant portion of the cells in a short time. In the Ea.hy926 culture, a decrease in the rate of proliferation was observed, but throughout the experiment the positive dynamics of cell culture growth continued. In the presence of the extract, the absence of proliferating cells in the fibroblast culture and a decrease in their number in the Ea.hy926 culture were noted.</p></sec><sec><title>Conclusion</title><p>Conclusion. Experimental conditions, test culture and evaluation methods must be selected individually based on the characteristics of the material being tested.
