положения</p><p>Доксорубицин – антрациклиновый химиотерапевтический препарат, который останавливает пролиферацию, активирует апоптоз и обладает кумулятивной и дозозависимой кардиотоксичностью. В основе кардиотоксичности антрациклинов лежит повреждение клеток миокарда, сопровождающееся дисфункцией левого желудочка. Дисфункция теломер играет важную роль в процессах клеточного старения, которые ассоциированы с повреждением миокарда.</p></sec><sec><title> </title><p> </p></sec><sec><title>Цель</title><p>Цель. Оценить экспрессию генов, кодирующих белки, вовлеченные в процесс регуляции длины теломерных участков ДНК кардиомиоцитов крыс линии Wistar в условиях субхронического низкодозового воздействия доксорубицина.</p></sec><sec><title>Материалы и методы</title><p>Материалы и методы. В исследование включены две группы самцов крыс линии Wistar: экспериментальная группа (10 крыс, еженедельная инъекция доксорубицина в хвостовую вену в дозе 2 мг/кг в течение 4 недель) и контрольная группа (10 крыс, еженедельная инъекция 0,9% NaCl в хвостовую вену в течение 4 недель). С помощью ПЦР определен уровень экспрессии гена Tert и генов шелтеринов Trf1, Trf2, Rap1, Tin2, Tpp1 и Pot1.</p></sec><sec><title>Результаты</title><p>Результаты. В экспериментальной группе наблюдалось повышение экспрессии генов шелтеринов Trf1, Trf2, Tpp1 и Tin2 по сравнению с контролем. Уровень мРНК генов Pot1 и Rap1 был снижен в экспериментальной группе относительно контрольных животных. Ген Tert в изученных группах экспрессировался на крайне низком уровне.</p></sec><sec><title>Заключение</title><p>Заключение. Установлено, что удлинение теломерных участков ДНК кардиомиоцитов крыс линии Wistar в ответ на субхроническое низкодозовое воздействие доксорубицина обусловлено изменениями в профиле экспрессии генов, кодирующих белки, ответственные за поддержание гомеостаза теломер, и, вероятно, происходит за счет запуска механизма альтернативного удлинения теломер.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>Highlights</title><p>Highlights</p><p>Doxorubicin is an anthracycline chemotherapeutic drug that stops proliferation, activates apoptosis, and has cumulative and dose–dependent cardiotoxicity. The cardiotoxicity of anthracyclines is based on damage to myocardial cells, accompanied by left ventricular dysfunction. Telomere dysfunction plays an important role in cellular aging processes that are associated with myocardial damage.</p></sec><sec><title> </title><p> </p></sec><sec><title>Aim</title><p>Aim. To evaluate the expression of genes encoding proteins involved in the regulation of the length of telomeric DNA regions of Wistar rat cardiomyocytes under subchronic low-dose doxorubicin exposure.</p></sec><sec><title>Methods</title><p>Methods. The study included two groups of male Wistar rats: an experimental group (10 rats, weekly injection of doxorubicin into the tail vein at a dosage of 2 mg/kg for 4 weeks) and a control group (10 rats, weekly injection of 0.9% NaCl into the tail vein for 4 weeks). The expression level of TERT and shelterin genes Trf1, Trf2, Rap1, Tin2, Tpp1 and Pot1 was determined using PCR.</p></sec><sec><title>Results</title><p>Results. The experimental group was characterized by increased expression of the shelterin genes Trf1, Trf2, Tpp1 and Tin2 compared to the controls. The mRNA level of Pot1 and Rap1 was lower in the experimental group compared to the controls. The TERT gene was expressed at an extremely low level.</p></sec><sec><title>Conclusion</title><p>Conclusion.
