положения</p></sec><sec><title> </title><p> </p></sec><sec><title>Цель</title><p>Цель. Сравнительный анализ профиля генной и белковой экспрессии в первичных эндотелиальных клетках атерочувствительной коронарной артерии (ЭК-КА) и атерорезистентной внутренней грудной артерии (ЭК-ВГА), в том числе при их дисфункции.</p></sec><sec><title>Материал и методы</title><p>Материал и методы. В работе были применены полнотранскриптомное секвенирование и ультравысокоэффективная жидкостная хроматография с тандемной масс-спектрометрией. При биоинформатическом анализе данных полнотранскриптомного секвенирования оценивали кратность изменения экспрессии и количество транскриптов генов интереса на миллион прочтений. Анализ данных хромато-масс-спектрометрического анализа проводили при помощи ранжирования 2 986 детектированных молекул по уровню их экспрессии (определяемой по площадям хроматографических пиков).</p></sec><sec><title>Результаты</title><p>Результаты. Дисфункциональные ЭК отличались выраженным снижением экспрессии ряда генов маркеров эндотелиального фенотипа, компонентов базальной мембраны и субэндотелиального внеклеточного матрикса, а также преимущественным снижением экспрессии генов сигнальных путей ангиогенеза. Дисфункция ЭК также приводила к повышению экспрессии генов сигнальных путей окислительного и эндоплазматического стресса. В сравнении с ЭК-ВГА ЭК-КА характеризовались повышенной экспрессией белков сигнальных путей ангиогенеза, в том числе эфриновых рецепторов (EPHB2 и EPHB4), а также обеих субъединиц эндотелиального интегрина αvβ3 (ITGAV и ITGB3). ЭК-ВГА отличались повышенной экспрессией транскрипционных факторов эндотелиальной дифференцировки (ERG и FLI1), ангиопоэтинов (ANGPT2 и ANGPTL2), их рецепторов (PTPRB и TEK) и ко-рецепторов к VEGF (NRP1 и NRP2), а также более высокой экспрессией компонентов базальной мембраны и внеклеточного матрикса, гиперэкспрессированных в интактных ЭК (ламинина, коллагена IV типа, фактора фон Виллебранда, ангиопоэтиноподобного белка, бигликана, мультимерина-1 и катепсина С).</p></sec><sec><title>Заключение</title><p>Заключение. Дисфункциональные ЭК характеризуются сниженной активностью синтеза компонентов базальной мембраны и внеклеточного матрикса. ЭК-КА и ЭК-ВГА имеют выраженные различия в экспрессии маркеров эндотелиального фенотипа, белков базальной мембраны, внеклеточного матрикса и сигнальных путей ангиогенеза.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>Highlights</title><p>Highlights</p></sec><sec><title> </title><p> </p></sec><sec><title>Aim</title><p>Aim. To compare the gene and protein expression profile in primary human coronary artery endothelial cells (HCAEC) and human internal thoracic artery endothelial cells (HITAEC) in physiological and dysfunctional states.</p></sec><sec><title>Methods</title><p>Methods. To perform an unbiased and high-throughput analysis, we applied RNA sequencing and ultra-high performance liquid chromatography-tandem mass spectrometry. Fold change and transcripts per million were used as metrics to evaluate RNA sequencing results. Mass spectrometry data were assessed by ranking the expression of 2,986 detected molecules after measuring areas of respective chromatographic peaks.</p></sec><sec><title>Results</title><p>Results. Dysfunctional ECs showed decreased expression of the genes encoding endothelial phenotype markers, basement membrane components, extracellular matrix proteins, and angiogenic molecules, concurrently having higher expression of the genes encoding oxidative and endoplasmic reticulum stress proteins. In comparison to HITAEC, HCAEC had higher expression of angiogenic proteins (including B-type ephrin receptors EPHB2 and EPHB4) and both subunits of endothelial integrin αvβ3 (ITGAV and ITGB3). In turn, HITAEC had higher expression of endothelial transcription factors ERG and FLI1, angiopoietins (ANGPT2 and ANGPTL2) and their receptors (PTPRB and TEK), VEGF co-receptors (NRP1 и NRP2), and also higher expression of basement membrane and extracellular matrix components which have been overexpressed in the intact ECs (such as laminin, type IV collagen, von Willebrand factor, angiopoietin-related protein 2, biglycan, nultimerin 1, cathepsin С, and ADAMTS4).</p></sec><sec><title>Conclusion</title><p>Conclusion. Dysfunctional ECs have lower expression of basement membrane and extracellular matrix components.
