Определить ключевые белки и сигнальные пути, вовлеченные в патогенезе ревматической болезни сердца (РБС).</p></sec><sec><title>Материалы и методы</title><p>Материалы и методы. С помощью безметочного протеомного профилирования посредством высокоэффективной жидкостной хроматографией с масс-спектрометрией проведено протеомное профилирование фрагментов створок нативных клапанов сердца, эксплантированных у пациентов с РБС (n = 3). В качестве группы сравнения использованы фрагменты створок нативных клапанов сердца, пораженные кальцинирующим аортальным стенозом (КАС) (n = 5).</p></sec><sec><title>Результаты</title><p>Результаты. В тканях нативных клапанов сердца пациентов с РБС выявлен 81 уникальный белок, участвующий в организации внеклеточного матрикса и коллагеновых фибрилл, адгезии клеток, регуляции дифференцировки остеобластов, миграции клеток и пролиферации эндотелиальных клеток. Кроме того, при сравнении с нативными клапанами сердца, пораженными КАС, в нативных клапанах сердца, эксплантированных у пациентов с РБС, выявлено 33 дифференциально-экспрессированных белка, 24 из которых были гиперэкспрессированны, а 9 – гипоэкспрессированны.</p></sec><sec><title>Заключение</title><p>Заключение. В результате проведенного протеомного профилирования нативных клапанов сердца, эксплантированных у пациентов с РБС, выявлены уникальные и дифференциально-экспрессируемые относительно клапанов сердца, пораженных КАС, белки и сигнальные пути, вовлеченные в развитие данного патологического состояния. Полученные пилотные данные вносят вклад в понимание фундаментальных механизмов, лежащих в основе патогенеза РБС, а также могут быть использованы для поиска таргетных молекулярных мишеней для коррекции данной патологии.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>Highlights</title><p>Highlights</p></sec><sec><title> </title><p> </p></sec><sec><title>Aim</title><p>Aim. To identify key proteins and signaling pathways involved in the pathogenesis of rheumatic heart disease (RHD).</p></sec><sec><title>Methods</title><p>Methods. Using high-performance liquid chromatography-mass spectrometry, we performed label-free proteomic profiling of native heart valves explanted from RHD patients (n = 3). Native heart valves affected by calcific aortic valve disease (CAVD) (n = 5) were used as a comparison group.</p></sec><sec><title>Results</title><p>Results. 81 unique proteins involved in the organization of extracellular matrix and collagen fibrils, cell adhesion, the regulation of osteoblast differentiation, cell migration, and endothelial cell proliferation were identified in native heart valves obtained from RHD patients. Furthermore, when compared with native heart valves affected by CAVD, 33 differentially expressed proteins were identified in native heart valves affected by RHD (24 upregulated and 9 downregulated).</p></sec><sec><title>Conclusion</title><p>Conclusion. Proteomic profiling of native heart valves explanted from RHD patients revealed unique and differentially expressed proteins and signaling pathways involved in the development of this pathological condition relative to heart valves affected by CAVD.
